Getting sick is something that people cannot avoid. There are many types of diseases clinically, and more serious diseases still need to be diagnosed through some medical examinations. For example, some diseases also require pathological sections, but most people do not know what pathological sections are, so it is easy to make some misunderstandings when making pathological sections. It is recommended to learn more about its nature and significance. 1. Pathological sections Pathological section refers to taking a certain size of diseased tissue and making it into pathological sections using pathological histological methods [usually the diseased tissue is embedded in a paraffin block, cut into thin slices with a slicer, and then stained with hematoxylin-eosin (HE)], and the lesion is further examined under a microscope. The occurrence and development process of the lesion is followed, and finally a pathological diagnosis is made. 2. Source The method of tissue sampling is an important procedure for making slices. Tissue sampling is obtained from the human body (surgical resection specimens, biopsy specimens, autopsy specimens) or animals according to the specific requirements of teaching, scientific research and external examination, and the location and method of sampling are determined. The sample collector needs to master the basic theoretical knowledge of anatomy, histology, and pathology, as well as the practical operation techniques. Each tissue and organ has a specific location and method for sampling, and tissues cannot be randomly cut as slide materials. 3. Pay attention to dyes The HE staining procedure is as follows: dewaxing, debenzening, rehydration, staining, dehydration, transparency, and sealing. The contrast stain in conventional hematoxylin staining is eosin. In recent years, some laboratories in the UK and the US have used phloxine. In addition, Orange G, Biebrich scarlet, Bordeaux red, etc. are also used as contrast stains. Eosin is a commonly used dye for staining cytoplasm, collagen fibers, muscle fibers, eosinophilic granules, etc. |
